海胆酮是一种酮式类胡萝卜素,主要从海胆及藻类等海洋生物中提取。本文研究海胆酮对乙酰胆碱酯酶(acetylcholinesterase,AChE)的抑制作用,应用酶动力学、荧光光谱、圆二色光谱和分子对接技术研究海胆酮对AChE的抑制机理,并用淀粉样β蛋白片段25~35(amyloid beta-peptide 25-35,Aβ25-35)诱导大鼠肾上腺嗜铬细胞瘤细胞(PC12细胞)建立阿尔茨海默症(Alzheimer’s disease,AD)模型,研究海胆酮对AD细胞模型氧化应激损伤的作用。结果表明,海胆酮有很强的AChE抑制活性,其半抑制质量浓度为(16.29±0.97)μg/mL,抑制常数Ki为3.82 μg/mL,表现为竞争性抑制;海胆酮可诱导AChE二级结构改变,更容易与AChE活性中心氨基酸Ser200、His440、Trp84和Tyr121结合,阻碍底物碘代硫代乙酰胆碱(acetylthiocholine iodide,ATCI)与酶结合,从而引起酶活力降低。海胆酮能有效抑制Aβ25-35诱导PC12细胞的AChE活力,降低丙二醛含量,增加超氧化物歧化酶、过氧化氢酶和谷胱甘肽过氧化物酶活力,减轻Aβ25-35诱导的PC12细胞氧化应激损伤。本研究基于AChE和氧化应激阐明了海胆酮对AD的潜在作用机制,为海胆酮在功能食品、生物医药等领域的应用提供了数据支持和理论根据。  相似文献

The deterministic and probabilistic prediction of ship motion is important for safe navigation and stable real-time operational control of ships at sea. However, the volatility and randomness of ship motion, the non-adaptive nature of single predictors and the poor coverage of quantile regression pose serious challenges to uncertainty prediction, making research in this field limited. In this paper, a multi-predictor integration model based on hybrid data preprocessing, reinforcement learning and improved quantile regression neural network (QRNN) is proposed to explore the deterministic and probabilistic prediction of ship pitch motion. To validate the performance of the proposed multi-predictor integrated prediction model, an experimental study is conducted with three sets of actual ship longitudinal motions during sea trials in the South China Sea. The experimental results indicate that the root mean square errors (RMSEs) of the proposed model of deterministic prediction are 0.0254°, 0.0359°, and 0.0188°, respectively. Taking series #2 as an example, the prediction interval coverage probabilities (PICPs) of the proposed model of probability predictions at 90%, 95%, and 99% confidence levels (CLs) are 0.9400, 0.9800, and 1.0000, respectively. This study signifies that the proposed model can provide trusted deterministic predictions and can effectively quantify the uncertainty of ship pitch motion, which has the potential to provide practical support for ship early warning systems.  相似文献

A new, experimental method based on air flow rate rather than current is presented to optimize operating parameters for the stacks and systems of proton exchange membrane fuel cells (PEMFCs) for maximizing their net power. This approach is illustrated for a commercial 18 kW PEMFC module. The impact of contamination pressure drop across the cathode air filter is also investigated on the compressor behavior. It is further shown that a 4V reduction in the compressor voltage reduces its power consumption by 9.1%. Using the 3D graphs of the power-pressure-flow data, it is found that the stack pressure of 180 kPaa is superior to the higher tested pressures as it enhances the net power by 7.0 and 13.7% at different conditions. Application of the present study will lead to the development of PEMFCs with higher power output by optimizing stack pressure, stoichiometry and air flow to properly deliver the system design specifications.  相似文献

In this present work, the effect of lanthanum oxides (La2O3) on the thermal cycle behavior of TBC coatings and mechanical properties such as adhesion strength and microhardness of 8% Yttria Stabilized Zirconia (8YSZ) TBCs were investigated. CoNiCrAlY and aluminium alloy (Al–13%Si) were used as bond coat and substrate materials. 8YSZ and different wt % of La2O3 (10, 20, and 30%) top coatings were applied using the atmospheric plasma spray (APS) method. The thermal cycling test for TBC coated samples were conducted at 800 °C in the electric furnace. The XRD pattern shows that the La2O3 doped 8YSZ material transformed to cubic pyrochloric structured La2Zr2O7 during thermal cycling. Further, the Taguchi-based grey relation analysis (GRA) method was applied to optimize the TBC coating parameters to achieve better mechanical properties such as adhesion strength and microhardness. And the optimized La2O3/8YSZ TBC coating was coated on CRDI engine combustion chamber components. The engine was tested with microalgae biodiesel and hydrogen, and the results were promising for the TBC-coated engine. The engine performance increased while using La2O3/8YSZ coated components, and the emissions from engine exhaust gas such as CO, HC, and smoke reduced considerably. It was found that there was no separation crack and spallation of the coating layer in the microstructure. Ultimately, the microstructural analysis of the optimized TBC coated piston sample after 50 h of running in the diesel engine confirmed that the developed coating had a superior thermal insulation effect and longer life.  相似文献

The low performance of open-cathode proton-exchange-membrane fuel cells (OCPEMFCs) is attributed to the low-humidity ambient air supplied to the cathode using electric fans. To improve the OCPEMFC performance, this paper proposes a novel humidification method by collecting water purged from the anode and supplying it to the open cathode. The OCPEMFC performance is evaluated at various humidifier distances from the cathode inlet, and it is compared with that where no humidifier is used when the OCPEMFC operates under three different current levels of 1, 5, and 8 A. The results show that the novel design improves the stack power, and optimal performance is achieved at a humidifier distance of 2 cm. The energy efficiency achieves an improvement between 1.4% and 1.8% when a humidifier is used.  相似文献

For proton-exchange membrane fuel cells, the distribution of reactant flow in the stack is critical to the fuel cell's efficiency. The uneven distribution of reactant flow in the stack may cause poor current density, low performance, and material degradation. To understand and accurately predict the flow field in the proton-exchange membrane fuel cell system, the present study aims to develop a simple correlation to analyze the pressure drop in fuel cell stacks. The flow channel in each cell of a stack is treated as a porous medium, and a power-law model is used to approximate the porous medium momentum source term. For the stacks with fewer cell numbers, namely, 1, 5, and 10 cells, the parameters in the power law are established based on the experimental data. Then, a correlation is developed to simulate the flow and predict the pressure drop in the stack with higher cell numbers (ie, 20 and 40 cells). The simulations show that the pressure drop in each cell of a stack is almost invariable, and the average pressure drop decreases with increasing the number of cells. The flow uniformity in the stacks with different cell numbers is evaluated using the dimensionless pressure drop and the pressure drop ratios. It suggests that the lower the cell number, the more uniform the pressure drop. The developed model is conducive to efficiently designing the flow channel for a fuel cell stack with large cell numbers.  相似文献

为明确大豆蛋白纳米纤维的结构形成和扩宽铁强化剂的食品工业应用,以大豆分离蛋白(soy protein isolate,SPI)为原料,通过5 h的酸热处理制备纳米纤维(soy protein isolate fibrils,Fib SPI),系统研究纤维形成前后蛋白结构的变化,并进一步制备铁纳米颗粒(iron nanoparticles,Fe NPs),探究Fib SPI对铁的稳态化作用。研究结果表明:在酸热处理过程中,SPI产生大量的β-折叠结构,其与硫磺素T结合,显示出增强的荧光强度;此外,7S组分先发生降解,利于纤维成核形成,随后11S逐渐被水解,促进纤维生长;同时水解产生大量的小肽组分,提高了产物的还原力。研究进一步利用Fib SPI递送铁纳米颗粒(Fe NPs),发现与原始SPI相比,铁纳米颗粒可在Fib SPI原位形成胶体稳定的铁-大豆蛋白纳米纤维复合物(Fe FibSPI),并以Fe(II)形式存在,其对乳液体系色泽及稳定性的影响较硫酸亚铁或氯化铁小。该研究可为构建新型植物基铁强化剂递送体系提供理论和方法指导。  相似文献

The oxygen starvation in fuel cells is an important reason for the deterioration of durability. The segmented fuel cell is a method to study the gas distribution inside the fuel cell. In order to study the influence of the grooving method on segmented fuel cell and its application in oxygen starvation diagnosis, a five-serpentine-channel three-dimensional two-phase simulation model is established by FLUENT. Through steady-state simulation, the effect of grooving method on fuel cell performance is studied. The overall performance (polarization curve) of the fuel cell drops slightly, but the current density distribution on the anode graphite plate changes greatly due to the grooves. The “current concentration” phenomenon is proposed based on the current density distribution. Through dynamic simulation, the oxygen starvation under current load mode and voltage load mode is simulated, and the “starvation coefficient” is defined as an oxygen starvation diagnostic index. In the current load mode, the “starvation coefficient” never exceed 15%, because when the oxygen starvation is severe, the simulation cannot converge or even cannot maintain, which corresponds to the voltage reversal in reality. However, in the voltage load mode, the “starvation coefficient” can reach up to 100%. The conclusions have important guiding significance for the judgment of the internal reaction uniformity of the segmented fuel cell by grooving method and provide a theoretical basis for judging whether a fuel cell is out of oxygen by segmented fuel cell.  相似文献

This study represents the results of the analysis and optimization of an integrated system for cogenerating electricity and freshwater. This setup consists of a Solid Oxide Fuel cell (SOFC) for producing electricity. Unburned fuel of the SOFC is burned in the afterburner to increase the temperature of the SOFC's outlet gasses and operate a Gas turbine (GT) to produce additional power and operate the air compressor. At the bottom of this cycle, a combined setup of a Multi-Effect Desalination (MED) and Reverse Osmosis (RO) is considered to produce freshwater from the unused heat capacity of the GT's exhaust gasses. Also, a Stirling engine is used in the fuel supply line to increase the fuel's temperature. Using LNG and the Stirling engine will replace the fuel compressor with a pump which increases the system performance and eliminates the need for the expansion valve. To study the system performance a mathematical model is developed in Engineering Equation Solver (EES) program. Then, the system's simulated data from the EES has been sent to MATLAB to promote the best operating condition based on the optimization criteria. An energetic, exergetic, economic, and environmental analysis has been performed and a Non-dominated Sorting Genetic Algorithm (NSGA-II) is used to achieve the goal. The two-objective optimization is performed to maximize the exergetic efficiency of the proposed system while minimizing the system's total cost of production. This cost is a weighted distribution of the Levelized Cost of Electricity (LCOE) and Levelized Cost of freshwater (LCOW). The results showed that the exergetic and energetic efficiencies of the system can reach 73.5% and 69.06% at the optimum point. The total electricity production of the system is 99 MW. The production cost is 11.71 Cents/kWh, of which 1.04 Cents/kWh is emission-related and environmental taxes. The freshwater production rate is 42.44 kg/s which costs 4.38 USD/m3.  相似文献

The gas purging states affect electricity output and energy storage capacity of unitized regenerative fuel cells. In this study, a model of unitized regenerative fuel cell is established. Cell voltages and operating temperatures influences on the dynamic distribution of thermal fluid during purging process and the discharge of residual liquid water in electrolytic cell mode are investigated. The motivation of the present study is better understanding the gas purging characteristics and its effect on reaction behaviors of unitized regenerative fuel cells. Simulation results reveal a significant influence of purging gas temperature on the water flooding and a great effect of operating voltage on the water diffusion. The operating temperature of electrolytic cell model almost has little effect on purging results at different cell temperature and the same purging gas temperature. When the purging gas temperature is changed, higher temperatures of cell and purging gas facilitate liquid water discharging out from the cell regions. In cell water flooding situation, when having large liquid content, the purging gas has little effects on the water expelling process.  相似文献
